In Transport Fever it was tested and the conclusion was that looking at the graphs the longer multi-stop lines tended to generate overall larger passenger numbers, but from a strictly profit standpoint, 2-stop point-to-point lines were overall more profitable, because you could tailor the capacity to the demand.
